Immunofluorescence of anti-Non-fucosylated xyloglucan-5 antibody CCRC-M48

Localizationofnon-fucosylatedxyloglucan-5(red)inArabidopsisthalianahypocotyl,CalcufluorWhitecounterstain(blue)andcellwallautofluorescence(yellow).

The31days-oldhypocotylswereimmersedin150μLPMEfixationbuffer(25mMPIPES,1mMMgSO4,1mMEGTA)andthensubjectedtothreeconsecutivecyclesof5min-longvacuuminfiltration(21°C,68kPa).AfterwardstheywerewashedthreetimesinPME(21°C,68kPa)priortostorageat4°CinPME.Hypocotylswereencasedin1cm3blocksof5%agarat65°C,andstoredat4°Ctoset.Transverse40μmthicksectionswerecutfromsegmentsusingaVT100Svibratingmicrotome(Leica)andblockedforatleast1hin5%bovineserumalbumininTBST.Blockingsolutionwasdiscardedandsectionswereincubatedat4°Cfor16hwith5μloftheanti-Non-fucosylatedxyloglucan-5antibody,followedby2washesin100μLTBST.Sectionswerethenincubatedfor1hat21°Cinthedarkin10μlof2μg/μlAlexaFluorTM568donkeyanti-mouseIgG(H+L;1:36).Sectionswereagainwashedtwicein40μLTBSTpriortocounter-stainingwith0.015%CalcofluorWhite(Sigma-Aldrich).Sectionswereagainwashedtwicein100μLTBSTtoremoveexcesscounter-stainandunboundsecondaryantibody.ImmunofluorescenceofAlexaFluor568wasexcitedwitha561nmlaser,andemittedlightfilteredat575–600nm,whileCalcufluorWhitewassubsequentlyscannedonanindependentchannelwitha405nmlaserandemissionobservedat420–430nmusinglaserscanningmicroscopeZeissLSM780point-scansystemat1024×1024pixels(pixelsize,0.6–0.83μm)witha10Xobjective.
